Representative two-color flow cytometry analysis of CD8+ T cells purified by positive isolation from PBMC. Cells were labeled with monoclonal antibodies to CD4 [phycoerythrin (PE)] and CD8 [fluorescein isothiocyanate (FITC)]. The purity of isolated CD8 fractions was assessed after gating on lymphocytes based on their forward- and side-scatter characteristics. Collective patient data showed 99.36% ± 0.33% (mean±SD) of positively selected cells were CD8+, and 0.2% ± 0.11% (mean±SD) were single-positive for CD4.

Neighbor-joining tree reconstructed with HIV-1 sequences from all patients. Bootstrap values were calculated using 1000 replicates. All clones clustered in a patient-specific manner confirming the absence of PCR contamination.
